| Other names | AID, also known as Activation-induced cytidine deaminase or Activation induced deaminase, is a protein involved in the immune response. It is encoded by the AICDA gene in humans and is commonly referred to as AICDA_HUMAN. AID plays a crucial role in the generation of antibodies by B cells. Another name for AID is ARP2, or ARP 2. It is also referred to as CDA2 or CDA 2, which stands for Cytidine aminohydrolase. AID is integrated into Burkitt's lymphoma cell line Ramos, a cell line derived from a tumor commonly found in the jaw or abdomen. Activation-induced cytidine deaminase operates by deaminating cytidine residues in DNA, resulting in the formation of uracil. This process is called somatic hypermutation and is responsible for the introduction of genetic diversity in the variable region of immunoglobulins. AID is also involved in class switch recombination, which allows B cells to switch the type of antibody they produce. Mutations in the AICDA gene can lead to Hyper-IgM syndrome type 2 (HIGM2), a rare immune disorder characterized by high levels of circulating IgM and low levels of other antibody types. Defects in AID function impair somatic hypermutation and class switch recombination, leading to an impaired immune response. In summary, Activation-induced cytidine deaminase, or AID, is a key player in the immune response, responsible for genetic diversification of antibodies. It is encoded by the AICDA gene and is involved in somatic hypermutation and class switch recombination. Mutations in AID can result in Hyper-IgM syndrome type 2, causing immune system dysfunction. Furthermore, AID is integrated into the Burkitt's lymphoma cell line Ramos, contributing to its pathogenesis. |

Background | Single-stranded DNA-specific cytidine deaminase. Involved in somatic hypermutation (SHM), gene conversion, and class-switch recombination (CSR) in B-lymphocytes by deaminating C to U during transcription of Ig-variable (V) and Ig-switch (S) region DNA. Required for several crucial steps of B-cell terminal differentiation necessary for efficient antibody responses (PubMed:18722174, PubMed:21385873, PubMed:21518874, PubMed:27716525). May also play a role in the epigenetic regulation of gene expression by participating in DNA demethylation (PubMed:21496894). |
| Tissue specificity | Strongly expressed in lymph nodes and tonsils. |
| Posttranslational modification | Ser-38 is the major site whereas Thr-27 is the minor site of phosphorylation. Phosphorylation regulates its class-switch recombination activity.Probably monoubiquitinated on several residues by RNF126. |
