Tris-EDTA Antigen Retrieval Solution(10×,pH9.0)

Antigen retrieval is an essential process in tissue production to address two common issues that can affect the immunohistochemical staining process. Firstly, certain chemical reagents can block the antigen, preventing its display. Secondly, heat can distort the peptide chain of the antigen, further hindering its visibility. By using specific buffers such as citrate, EDTA, or Tris under hot conditions, these antigens can be re-exposed without damaging the epitopes. This improves the detection rate of antigens, reduces background staining, and enhances diagnostic accuracy.
Tris-EDTA antigen retrieval solution (10x, pH9.0) is particularly effective in removing protein crosslinks caused by aldehyde fixatives. It fully exposes the epitopes in paraffin sections and other samples. This solution can be utilized for antigen retrieval in samples fixed with paraformaldehyde, formaldehyde, or other aldehyde reagents, including paraffin sections and frozen sections. Not only does antigen retrieval enhance the immunostaining effect of paraffin sections, but it can also improve the staining quality of frozen sections to varying degrees. In cases where the immunostaining effect of frozen sections is unsatisfactory, antigen retrieval can be considered. With each film requiring 10ml of antigen retrieval solution (1x), the 10x antigen retrieval solution can accommodate antigen retrieval for 100 samples.

Solution

It is recommended to explore the optimal heating time for various samples and target proteins after soaking them in antigen repair solution (1×).
