Description | SLP-76 was initially identified as a ZAP-70 protein tyrosine kinase substrate after T cell receptor (TCR) engagement in Jurkat leukemic T cells. The SLP-76 gene locus is localized on chromosome 5q33 in humans, and partially characterized in mice. The human and murine cDNAs both code for 533 amino acid proteins that share 72% identity and consist of three modular domains. The N-terminal domain contains an acidic region with a PEST domain and several phosphorylated tyrosine residues following TCR engagement. SLP-76 also features a central proline-rich domain and a C-terminal SH2 domain. Other proteins have been identified that bind to SLP-76 constitutively or after receptor engagement, indicating that SLP-76 acts as an adaptor or scaffold molecule. Experiments using SLP-76 deficient T cell lines or mice have shown that SLP-76 has a positive impact on T cell development and activation, as well as mast cell and platelet function. |
