Background

Caspase-11 (also known as inflammatory caspase) is a crucial component of the NLRP3 inflammasome, which is responsible for activating CASP1 and inducing the secretion of IL1B and IL18. It is primarily activated by non-canonical triggers such as UVB radiation, cholera enterotoxin subunit B, and cytosolic LPS.
Aside from its role in the NLRP3 inflammasome, caspase-11 also promotes pyroptosis (a form of cell death) and the release of IL1A, IL18, and HMGB1 in response to non-canonical inflammasome activators. This process involves the cleavage and activation of GSDMD.
Furthermore, caspase-11 plays a critical role in limiting the replication of Salmonella typhimurium in colonic epithelial cells during infection. In the later stages of the infection, cytosolic Salmonella LPS triggers caspase-11 activation, leading to pyroptosis of infected cells, release of IL18, and extrusion of infected cells into the gut lumen. This process helps restrict bacterial replication and promotes mucosal inflammation by triggering cytokine secretion and immune cell activation.
Additionally, caspase-11 is involved in LPS-induced IL6 secretion, which may not necessarily require its enzymatic activity. It is also implicated in cell death induced by endoplasmic reticulum stress and by cytotoxic APP peptides found in Alzheimer's patient brains.
Caspase-11 can be directly activated by binding to LPS without the need for an upstream sensor. It does not directly process IL1B but is involved in the regulation of antiviral innate immune activation through the cleavage of MB21D1.
