Description

This particular gene is responsible for producing a transcription factor, which has the ability to activate specific genes involved in the unfolded protein response (UPR) during times of endoplasmic reticulum (ER) stress. Interestingly, this protein is synthesized as a transmembrane protein, which means it's embedded within the ER. Its role revolves around sensing and reacting to ER stress, and when ER stress initiates protein turnover, it functions as a transcription factor in the nucleus through an ER stress response element (ERSE) present in genes that encode chaperones for the ER. In addition, it's been discovered that this gene is pivotal in maintaining the survival of quiescent cells of squamous carcinoma, although it does not have an impact on proliferative cells. There's been some debate regarding whether polymorphisms in this gene might be correlated with diabetes in certain populations, but one particular polymorphism has been linked to increases in plasma cholesterol levels. Additionally, researchers are currently investigating whether this gene may in fact be a valuable therapeutic target for individuals afflicted with cystic fibrosis.

Other namesATF6, also known as activating transcription factor 6, is a crucial protein involved in cellular stress response. Specifically, ATF6 alpha is a variant of ATF6 that plays a significant role in regulating gene expression under various conditions. This transcription factor is activated in response to endoplasmic reticulum (ER) stress, a condition where misfolded proteins accumulate in the ER.
When unfolded or misfolded proteins accumulate in the ER, ATF6 is released from its inactive state and translocates to the Golgi apparatus. In the Golgi, ATF6 undergoes proteolytic cleavage by specific enzymes, resulting in the production of its active form, ATF6 alpha. This processed ATF6 alpha then translocates to the nucleus, where it functions as a transcription factor.
Once in the nucleus, ATF6 alpha binds to specific DNA sequences known as ER stress response elements (ERSEs) present in the promoter regions of target genes. By binding to these ERSEs, ATF6 alpha activates the transcription of various genes involved in ER protein folding, ER-associated degradation, and overall cellular adaptation to ER stress.
The activity of ATF6 alpha is regulated by the levels of ER stress. Under normal conditions, ATF6 alpha remains in an inactive state. However, when ER stress occurs, ATF6 alpha is activated and initiates the unfolded protein response (UPR), which aims to restore ER homeostasis. The UPR includes mechanisms such as increasing ER protein folding capacity and reducing protein synthesis to alleviate ER stress.
Overall, ATF6 alpha is an essential transcription factor that plays a pivotal role in the cellular stress response by regulating the expression of genes involved in ER protein homeostasis. Its activation and subsequent modulation of gene expression are critical for cell survival and adaptation to ER stress.

ApplicationThe recommended dilution for Western blotting is between 1:500 and 1:2000. For immunohistochemistry, the suggested dilution range is from 1:50 to 1:200. And if you are using ELISA with a peptide, it is advised to dilute at a range of 1:20000 to 1:40000. It is important to keep the dilution within these ranges for optimal results in each respective assay.

Target

Background

Transmembrane glycoprotein of the endoplasmic reticulum that functions as a transcription activator and initiates the unfolded protein response (UPR) during endoplasmic reticulum stress. Cleaved upon ER stress, the N-terminal processed cyclic AMP-dependent transcription factor ATF-6 alpha translocates to the nucleus where it activates transcription of genes involved in the UPR. Binds DNA on the 5'-CCAC[GA]-3'half of the ER stress response element (ERSE) (5'-CCAAT-N9-CCAC[GA]-3') and of ERSE II (5'-ATTGG-N-CCACG-3'). Binding to ERSE requires binding of NF-Y to ERSE. Could also be involved in activation of transcription by the serum response factor. May play a role in foveal development and cone function in the retina.

Tissue specificityUbiquitous.
Posttranslational modificationDuring unfolded protein response, a fragment of approximately 50 kDa containing the cytoplasmic transcription factor domain is released by proteolysis. The cleavage seems to be performed sequentially by site-1 and site-2 proteases.N-glycosylated. The glycosylation status may serve as a sensor for ER homeostasis, resulting in ATF6 activation to trigger the unfolded protein response (UPR).Phosphorylated in vitro by MAPK14/P38MAPK.
