Description

By releasing the terminal His-Leu, Angiotensin Converting Enzyme (ACE) converts angiotensin I to angiotensin II, leading to an enhanced vasoconstrictor effect. Additionally, ACE possesses the ability to inactivate bradykinin, a potent vasodilator. Another function of ACE is its glycosidase activity, which involves cleaving the mannose linkage in the GPI moiety, thereby releasing GPI-anchored proteins from the membrane. It is important to note that the generated content is rearranged but contains the same information as the original text.

Other namesACE, also known as angiotensin converting enzyme, is a peptidyl dipeptidase that plays a crucial role in the renin-angiotensin-aldosterone system. It exists in multiple isoforms, including ACE T and ACE_HUMAN, and is expressed in various tissues such as the somatic and testis.
ACE converts angiotensin I to angiotensin II, a potent vasoconstrictor, and is also involved in the degradation of bradykinin, a vasodilator. It is thus a key player in regulating blood pressure and electrolyte balance.
Interestingly, ACE has also been found to have non-classical functions, such as being involved in the immune response and the breakdown of amyloid-beta plaques in Alzheimer's disease.
Overall, ACE is a highly important enzyme with multiple isoforms and functions that must be carefully regulated to maintain homeostasis in the body.
SourceRabbit
SpecificityThe ACE1 Antibody specifically targets and identifies total ACE1 levels in the body. The detection is based on the endogenous levels of ACE1, providing accurate results.

ApplicationThe recommended dilutions for western blotting range from 1:500 to 1:2000, while for immunohistochemistry they fall within the range of 1:50 to 1:200. If performing ELISA with peptide samples, the suggested dilution range is 1:20000 to 1:40000. It is important to note that these dilutions may require optimization for specific experiments and should be used as a starting point.

Target

Background

Converts angiotensin I to angiotensin II by release of the terminal His-Leu, this results in an increase of the vasoconstrictor activity of angiotensin. Also able to inactivate bradykinin, a potent vasodilator. Has also a glycosidase activity which releases GPI-anchored proteins from the membrane by cleaving the mannose linkage in the GPI moiety.

Tissue specificityUbiquitously expressed, with highest levels in lung, kidney, heart, gastrointestinal system and prostate. Isoform Testis-specific is expressed in spermatocytes and adult testis.
Posttranslational modificationPhosphorylated by CK2 on Ser-1299; which allows membrane retention.
Celluar localizationEndosome;Extracellular region or secreted;Lysosome;Plasma Membrane;

Western blot analysis of ACE1 expression in mouse kidney

AF5197 at 1/100 staining human lung tissue sections by IHCP. The tissue was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The tissue was then blocked and incubated with the antibody for 1.5 hours at 22°C. An HRP conjugated goat anti-rabbit antibody was used as the secondary.
