It interacts with different cyclins, namely cyclin E and cyclin A, to form functional kinase complexes that phosphorylate downstream targets. CDK2 phosphorylates various proteins including CTNNB1, USP37, p53/TP53, NPM1, CDK7, RB1, BRCA2, MYC, NPAT, EZH2. Its activities include triggering the duplication of centrosomes and DNA, promoting the E2F transcriptional program and initiation of DNA synthesis at the G1-S transition, and modulating G2 progression. CDK2 also controls the timing of entry into mitosis and meiosis by phosphorylating and activating cyclin B/CDK1. It coordinates the activation of cyclin B/CDK1 at the centrosome and nucleus. In human embryonic stem cells, CDK2 plays a crucial role in maintaining a balance between cellular proliferation, cell death, and DNA repair.
