Pregabalin

Lyrica, also known as pregabalin, is a medication that has multiple effects on the central nervous system. It is derived from gamma-amino butyric acid (GABA) and exhibits properties such as anti-convulsant, anti-epileptic, anxiolytic, and analgesic activities. The way pregabalin works is by selectively binding to alpha2delta (A2D) subunits of presynaptic voltage-dependent calcium channels (VDCCs) in the CNS.
By binding to these VDCC A2D subunits, pregabalin effectively blocks the influx of calcium into cells. This prevents the release of various neurotransmitters, including glutamate, norepinephrine, serotonin, dopamine, and substance P, from the hyperexcited neurons' presynaptic nerve terminals. Consequently, synaptic transmission is inhibited, and the overall excitability of neurons is reduced.
It is important to note that pregabalin does not directly bind to GABA-A or GABA-B receptors, nor does it interfere with the uptake or degradation of GABA. Its mechanism of action primarily revolves around its interaction with VDCC A2D subunits in order to modulate neuronal activity and neurotransmitter release.
