Interleukin-2 (IL-2) is a small protein with a molecular weight of 15.5kDa and composed of 133 amino acid residues. It possesses three cysteine amino acids at positions 58, 105, and 125. The intra-chain disulfide bond formed by cysteine 58 and cysteine 105 is crucial for maintaining the biological activity of IL-2. T cells or T cell lines are the primary producers of IL-2. This cytokine is typically released by mitogen-stimulated CD4-positive or CD8-positive T cell subsets. The secretion of IL-2 by CD4-positive T cells is mainly triggered by alloantigens, whereas T cells located in the spleen, lymph nodes, and tonsils produce IL-2 when stimulated.
 
IL-2 has shown promising results in preventing tumor formation and stopping the growth of existing tumors. In clinical trials, IL-2 has been found to stimulate PBMC and TIL to turn into LAK cells, which are known to show significant anti-tumor activity in vitro. Renal cell carcinoma, melanoma, non-Hodgkin's lymphoma, and colorectal cancer have been found to respond well to LAK/IL-2 treatment. However, the response to IL-2 treatment may vary for other types of cancers such as liver, ovarian, bladder, and lung cancer.

 

Furthermore, IL-2 gene therapy has been tested in clinical trials for melanoma, renal cell carcinoma, and neuroblastoma, and the results have been positive. Additionally, IL-2 has been found to have anti-viral properties, and it can enhance the antiviral activity of CTL and NK cells, as well as stimulate the production of IFN-γ to combat viral infections caused by immune deficiency.

Recently, IL-2 has also shown some promise in the treatment of active hepatitis and herpes simplex virus infection. Furthermore, it is being tested in the clinical validation of phase ⅱ for treating AIDS disease, nodular leprosy, mycobacterium tuberculosis infection, among others.
