To cultivate animal cells in vitro, the tissue of interest is first extracted from the animal and enzymatically treated with trypsin or collagenase to break down the extracellular matrix and dissociate individual cells. These individual cells are then seeded onto a substrate in a medium optimized for their growth and proliferation. With the right nutrients and conditions, the cells will continue to divide and form a monolayer, allowing researchers to study their biology and behavior in a controlled environment.
Primary culture is a process where tissues of animals are extracted from their bodies and subjected to enzymatic breakdown using trypsin, chelating agents like EDTA, or mechanical methods. These tissues are then dispersed into individual cells and cultivated in a suitable medium to ensure their survival, growth, and reproduction. To facilitate further growth, the cultured cells are subsequently divided and diluted into multiple vials, a procedure known as passage.

Cell culture in plant biotechnology involves the cultivation of easily dispersed tissues or callus in a controlled in vitro environment. To achieve cell proliferation and obtain large cell populations, the suspension cells are subjected to shock culture in liquid medium and subsequently dispersed into free cells. This allows for subculturing and the expansion of cell numbers.
